Functional gene polymorphisms of the molecules regulating inflammation

During last years a lot of data have been accumulated concerning the role of single nucleotide polymorphisms (SNP), forming specific alleles of certain genes, in the individual variations of immune reactivity. In this review we have summarized existing data on influence of the genetic functional polymorphisms of the early inflammatory genes responsible for the pathogen detection on the mode of defence reactions and predisposition to some illness. Common feature of the functional TLR gene polymorphisms considered to be a decrease in the detection and binding specific ligands and consequent decrease of pathogen-induced cell activation. It is evident that these results show only predisposition of individuals carrying some SNP to various diseases, but examples of certain allele linkage with mortality in sepsis and with atopy makes these investigations more than serious. Nowadays SNP studies are of especial importance because it can make prognosis for the risk of severe illness development, and from the other hand in advance propose specific individual therapy, including modern immunotherapy.
