The purpose of this study was to evaluate the level of 17 cytokines (IL-1p, TNFa, IFNy, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-10, IL-12 (p70), IL-13, IL-17, G-CSF, GM-CSF, MCP-1, MIP-ip) in serum and spontaneous and mitogen-induced (LPS, ConA) whole blood of healthy donors and to determine the normal values of human cytokine status. The marked inter-individual variation in serum level of IFNy, IL-13, IL-1 p, TNFa, MIP-1p, G-CSF and GM-CSF, and the significant negative age-related correlation with serum concentration of IFNy, TNFa, IL-12, IL-7 and GM-CSF were revealed. The serum levels of pro- inflammatory (IL-1p, TNFa), Th1 (IFNy) and Th2 cytokines (IL-13), as well as the growth factors (IL-7, GM-CSF) were found to be considerably decreased in older donors (> 43 years, mean 48) compared to healthy subjects at the age from 20 to 30 years (mean 24). It has been shown that the measurement of spontaneous and mitogen-stimulated cytokine production in whole blood cultures essentially up-grade the diagnostic value of cytokine status analysis. The obtained data of in vivo and ex vivo cytokines level, in detail presented as M ± S.E., median and min-max range, may be used as normal data-base in subsequent investigations.
